Das entsprechende Gen, SCP160, ist zwar nicht essentiell für das vegetative Zellwachstum; Mutanten, bei denen es verkürzt oder deletiert ist, zeigen jedoch verminderte Lebensfähigkeit, abnormale Morphologie (heterogene Zellgröße und Zellgestalt, teils schlauchförmig verlängerte Zellen, teils Tochterzellen ohne Zellkern; etwa 2 % der Mutanten besitzen zwei Kerne) und etwa zwei- bis dreifach erhöhten DNA-Gehalt. Darüberhinaus spalten bei Kreuzungen von Wildtypstämmen und Stämmen mit disruptiertem SCP160-Gen genetische Marker unregelmäßig auf, was darauf hinweist, daß Scp160p bei der korrekten Aufteilung der Chromosomen während der Zellteilung eine regulatorische Rolle spielen könnte. Das Vorhandensein von zehn potentiellen N-Glykosylierungsstellen in der Aminosäuresequenz und die Differenz zwischen der aus der Sequenz errechneten (134.7 kD) und der auf der SDS-PAGE beobachteten (etwa 160 kD) molekularen Masse von Scp160p legten den Schluß nahe, Scp160p sei glykosyliert. Ziel dieser Arbeit war nun, das Protein auf eine derartige posttranslationale Modifikation zu überprüfen. Darüberhinaus sollte festgestellt werden, ob Scp160p mit einem Membransystem assoziiert ist und die Lokalisation des Proteins in der Zelle sollte aufgeklärt werden. Eine N-Glykosylierung von Scp160p konnte mit Hilfe von drei Methoden, nämlich Behandlung mit PNGase F, Untersuchung von Scp160p mittels Glyco-Track Kit sowie Lektinaffinitäts- chromatographie ausgeschlossen werden. Fraktionierungsversuche zur Lokalisation von Scp160p innerhalb der Zelle ergaben, daß das Protein an ein Membransystem gebunden ist, von dem es durch Behandlung mit Salz gelöst werden kann, was darauf hinweist, daß die Membranassoziation über ionische Wechselwirkungen erfolgt. Bei der subzellulären Fraktionierung findet sich Scp160p sowohl in der Kernfraktion als auch im Überstand nach der Kernabtrennung, der die Mikrosomen, also Membranen des rauhen und glatten ER, enthält. Durch Immunfluoreszenzmikroskopie konnte bestätigt werden, daß Scp160p in der Kernmembran und im ER, das in Saccharomyces cerevisiae in engem Kontakt mit der Kernmembran steht, lokalisiert ist.
| Abstract in Englisch: | Scp160p, a protein from Saccharomyces cerevisiae, has been characterized and localized in the cell. The corresponding gene, SCP160, is not essential for vegetative cell growth, but shortening and distruption of the gene results in cells of decreased viability and abnormal morphology (heterogenous cell size and cell shape, hoseshaped cells and daughter-cells , which lack nuclei; about 2 % of the mutants contain two nuclei), the DNA content is increased two to three times. In addition, crossing of wildtype-strains and strains with disrupted SCP160-gene results in unregulatory distribution of genetic markers. This indicates, that Scp160p could play a regulatory role in the correct distribution of chromosomes during cell division. The aminoacid sequence of Scp160p contains ten potential N-glykosylation sides and a difference in molecular mass expected from the sequence (134.7 kD) and observed on SDS-PAGE (about 160 kD) suggested, Scp160p is glycosylated. In this work it was first to test the protein for this kind of posttranslational modifikation. Futhermore it should be determined, if Scp160p is bound to a membrenous system as well as the localization of the protein in the cell. N-glycosylation of Scp160p could be excluded by treatment with PNGase F, test with Glyco-Track Kit and lectin affinitychromatography. First cell fractionation experiments showed a membran-association of Scp160p, which can be solved by salt; this indicates, that Scp160p is bound to a membranous system in the cell by ionic interactions. In subcellular fractionation experiments Scp160p was found in the nuclear fraction as well as in the supernatent, which contains microsomes of rough and smooth ER-membranes. Indirect immunofluorescence microscopy confirmed, that Scp160p is a protein of the nuclear membran and the endoplasmatic reticulum, which is closely connected with the nuclear membrane in Saccharomyces cerevisiae.
